Immunoassay Intelligence

Previously known as ELISA Clinical Intelligence — now expanded into a comprehensive immunoassay intelligence framework

From optical density to clinical decision-making across ELISA, CLIA, and advanced immunoassays

This course moves beyond ELISA as a technique and develops a systems-level understanding of immunoassays as diagnostic tools. It integrates ELISA, CLIA, and modern immunoassay platforms with curve intelligence, analytical modeling, biological variability, and clinical decision-making.

Stop running assays. Start interpreting them — ELISA, CLIA, and the full immunoassay intelligence layer.

Overview

Immunoassay Intelligence — Built on ELISA Foundations

This course moves beyond ELISA as a technique and develops a systems-level understanding of immunoassays as diagnostic tools. It integrates ELISA, CLIA, and modern immunoassay platforms with curve intelligence, analytical modeling, biological variability, and clinical decision-making.

ELISA remains the foundational platform for understanding immunoassays. However, modern diagnostics extends beyond ELISA into multiple assay systems. This course has evolved from ELISA Clinical Intelligence into Immunoassay Intelligence, where ELISA serves as the analytical foundation for broader diagnostic interpretation.

Learners acquire both ELISA analysis competency and the interpretive reasoning to navigate ELISA vs CLIA discordance, immunoassay interference, and clinical decision logic — skills required in translational research and diagnostic laboratory settings.

Objectives

Learning Objectives

By the end of this immunoassay course, learners will demonstrate mastery across seven core competencies spanning ELISA interpretation, CLIA, analytical error intelligence, and clinical decision frameworks.

1

Differentiate immunoassay platforms (ELISA, CLIA, ECLIA) by analytical principle and clinical application

2

Perform and interpret ELISA curve fitting using 4PL/5PL models and goodness-of-fit metrics

3

Identify sources of analytical variability and interference in ELISA and immunoassay systems

4

Apply reference change values and biological variation data to immunoassay result interpretation

5

Detect hook effect, heterophile antibody interference, and matrix effects before clinical reporting

6

Integrate ELISA analysis and immunoassay data into structured clinical decision frameworks

7

Apply decision-level thinking to distinguish assay error from biological signal in diagnostic settings

Interactive Tool

EIA Engine™ — Immunoassay Intelligence Tool

Built on ELISA principles, expanded for full immunoassay analysis. A live analytical tool integrated into this course — not a simulation, but a real immunoassay calculator for your data.

EIA Engine™ — Immunoassay Intelligence Tool

PM Lab Suite · ELISA & Immunoassay Analytics

Live

Standard Curve

Input OD values and generate 4PL/5PL fitted curves with goodness-of-fit metrics

Interpolation

Calculate sample concentrations with confidence intervals and accuracy flags

Outlier Detection

Identify curve inconsistencies, hook effects, and poor-fit regions automatically

Error Propagation

Launch Tool
Curriculum

Seven Clusters, Fourteen Modules

A structured progression from ELISA foundations through immunoassay platform intelligence, analytical error mastery, and clinical decision systems.

Immunoassay Platforms Compared

ELISA, CLIA, ECLIA, and lateral flow — signal generation principles, detection limits, and clinical trade-offs across platforms.

ELISA Format Intelligence

Direct, indirect, sandwich, and competitive ELISA — selecting the right format and understanding its inherent analytical constraints.

Case-Based Learning

Case-Based Diagnostic Lab

Learners work through real immunoassay datasets — using ELISA interpretation skills, ELISA curve analysis, and ELISA vs CLIA reasoning to differentiate assay error from biological signal in clinical contexts.

IL-33

IL-33 ELISA Interpretation Across Patient Groups

Differentiating biological signal from assay noise in allergic and inflammatory conditions — a core ELISA interpretation challenge requiring curve-level and clinical-level analysis.

HMGB1

HMGB1 Immunoassay Error Intelligence

Haemolysis interference, matrix effects, and distinguishing active vs passive HMGB1 release — when ELISA analysis reveals pre-analytical contamination before clinical reporting.

Curve

ELISA Curve Distortion — 4PL Failure Scenarios

Hook effect detection, poor 4PL/5PL goodness-of-fit, and parallelism failure in ELISA curve fitting — identifying distortions before they propagate to clinical decisions.

CLIA

ELISA vs CLIA Discordance in Clinical Practice

When ELISA and CLIA yield discordant results for the same analyte — platform-specific interference, calibration differences, and the decision framework for resolving clinical ambiguity.

Framework

Structured Interpretation: The CLIP Layer

A four-step framework that transforms raw immunoassay data — from ELISA optical density to CLIA RLU — into clinically meaningful decisions. ELISA analysis is the entry point; clinical interpretation is the destination.

1

Validate Analytical Integrity

Confirm ELISA curve quality, control performance, and within-run precision. Identify hook effect, parallelism failure, or calibration drift before any clinical interpretation.

2

Assess Biological Plausibility

Is the immunoassay result biologically consistent? Check pre-analytical variables, matrix interference, and whether the result is within the expected physiological range for this patient.

3

Map to Disease Mechanism

Situate the biomarker within the relevant pathophysiological pathway. ELISA and immunoassay results gain meaning only when anchored to the disease biology they are intended to reflect.

4

Integrate with Clinical Context

Combine immunoassay data with clinical presentation, serial trends, and co-investigated analytes. A single immunoassay result rarely justifies a clinical decision in isolation.

CLIP Platform

Part of the ClinLab Intelligence Platform

This immunoassay course is part of CLIP — a connected ecosystem linking ELISA analysis, immunoassay interpretation, and clinical decision pathways. The learning path: Techniques (ELISA basics) → Immunoassay Intelligence → Lab Quality Systems.
